چکیده

It is shown that for carboxypeptidase Y catalyzed peptide synthesis the coupling yields using peptide esters as substrates and amino acids as nucleophiles are strongly dependent on the C-terminal amino acid residue. A dependence on the length of the acyl donating peptide chain is also observed, indicating the influence of residues other than the one directly involved in acylation of the enzyme. Using amino acid amides as nucleophiles, the yields are generally high (80 %-100 96). No peptide ester substrate with C-terminal D-amino acid residue can be used in this method of peptide synthesis since they are not substrates of the enzyme. However, the presence of Damino acids in the penultimate position does not prevent peptide synthesis. No dependence on the size of the leaving group of esterand depsipeptide substrates could be observed.

Recognition of C-terminal amide groups by (serine) carboxypeptidase Y investigated by site-directed mutagenesis.

Serine carboxypeptidases have the ability to hydrolyze peptides as well as peptide amides. Previously, it has been demonstrated that Asn51 and Glu145 (in the protonated form) each donate a hydrogen bond to the alpha-carboxylate of peptide substrate. It is here demonstrated by characterization of carboxypeptidase Y derivatives, mutationally altered at positions 51 and 145, that the same groups a...

Enzymatic Peptide Synthesis. Carboxypeptidase Y Catalyzed Formation of Peptide Bonds

It is demonstrated that carboxypeptidase Y from Saccharomyces cerevisiae can catalyze the formation of peptide bonds using N-acylamino acid esters as substrates and free amino acids or amino acid amides as nucleophiles, The coupling yields observed with free amino acids were max. 60 % for alanine and lysine and they depended strongly on the reaction parameters; viz. pH, temperature and concentr...

Aminoacyl-SNACs as small-molecule substrates for the condensation domains of nonribosomal peptide synthetases.

BACKGROUND Nonribosomal peptide synthetases (NRPSs) are large multidomain proteins that catalyze the formation of a wide range of biologically active natural products. These megasynthetases contain condensation (C) domains that catalyze peptide bond formation and chain elongation. The natural substrates for C domains are biosynthetic intermediates that are covalently tethered to thiolation (T) ...

Peptide bond formation on the ribosome: structure and mechanism.

The peptidyl transferase reaction on the ribosome is catalyzed by RNA. Pre-steady-state kinetic studies using Escherichia coli ribosomes suggest that catalysis (>10(5)-fold overall acceleration) is, to a large part, a result of substrate positioning, in agreement with crystal structures of large ribosomal subunits with bound substrate or product analogs. The rate of peptide bond formation is in...

Modulation of the Rate of Peptidyl Transfer on the Ribosome

The ribosome catalyzes peptide bond formationbetweenpeptidyl-tRNA in the P site and aminoacyl-tRNA in theA site. Here, we show that the nature of the C-terminal amino acid residue in the P-site peptidyl-tRNA strongly affects the rate of peptidyl transfer. Depending on the C-terminal amino acid of the peptidyl-tRNA, the rate of reaction with the small A-site substrate puromycin varied between 100...
